Regulation of histone acetylation on cell cycle and p53,p21~(WAF1) genes expression in malignant transformation of IEC-6

Objective To investigate the effects of histone acetylation on the expression of p53 and p21 WAF1 genes in malignant transformation of IEC-6. Methods The malignant transformation of IEC-6 was carried out by self-established three transition phases in which chemical cancerogenic agent MNNG and derivative PMA were used to the target cells for 6, 12, and 24 times. The transformants were cultured and the neucleoprotein of IEC-6 was extracted during every transition phase. The cell cycle distribution was studied by flow cytometry. The level of acetylated histones in chromatin associated with the p53 and p21 WAF1 was examined by chromatin immunoprecipitation (ChIP) assay. The expression of p53 and p21 WAF1 genes mRNA was detected by RT-PCR. Results Acetylation level of histone H3 in IEC-6 was higher after malignant transformation for 24 times than 6 times. Malignant transformation induced an accumulation of acetylated histone H3 in chromatin associated with p21 WAF1 gene. Expression of p53 and p21 WAF1 was detected weakly in 6 and 12 times malignant transformation of IEC-6. There was no expression of p21 WAF1 in malignant transformation of IEC-6 for 24 times. Conclusion In the malignant transformation of IEC-6 cell, the expression of p21 WAF1 and p53 genes is regulated by histone acetylation. The enhanced acetylation of histone H3 will restrain the expression of p21 WAF1 gene and further may affect the cell cycle of transformants.
