Inhibition effect on LPS-induced TNF-α release by homegrown and Australian Melaleuca alternifolia(Tea Tree)oil

Objective To compare the inhibition of the homegrown and Australian tea tree oil(TTO)solution on TNF-α release from RAW264.7 cells induced by LPS.Methods MTT assay was used to measure the effects of TTO on viability of RAW264.7 in vitro.RAW264.7 cells were pretreated with two kinds of TTO solution in the dose-and time-dependent manners,then stimulated by LPS.TNF-α in the supernate was tested by ELISA.Results In vitro,two kinds of TTO(0.001%)showed no cytotoxicity to RAW264.7 cells.Australian TTO(0.000 125%-0.001%)and homegrown TTO(0.000 25%-0.001%)significantly suppressed LPS-induced TNF-α release,respectively.0.000 25% Australian TTO pretreating for 3 h and homegrown TTO pretreating for 6 h showed significant protections against the stimulations of LPS,respectively.Conclusion In vitro,two kinds of TTO have the obvious inhibitory action,and there is no significant difference between Australian and homegrown TTO.
