Alkaline phosphatase,liver/bone/kidney,inhibits proliferation of ovarian cancer cell line SKOV3

Objective To investigate the expression of tissue-nonspecific alkaline phosphatase,liver/bone/kidney(ALPL) in human ovarian cancer cell lines,and determine its effect on the proliferation of ovarian cancer SKOV3 cells. Methods qRT-PCR was used to detect the expression of ALPL mRNA level in7 ovarian cancer cell lines(HEY,SKOV3,8910,ES-2,OVCAR3,3 AO and A2780). The results were compared with its level in normal ovarian tissues. After the SKOV3 cells(with low expression of ALPL) were interfered with lentiviral vector to induce ALPL overexpression,cell proliferation was measured by MTT assay and plate colony formation assay,and cell cycle were assessed by flow cytometry. Tumorigenesis was evaluated through xenograft growth of ALPL-overexpressed SKOV3 cells in nude mice. Results The expression level of ALPL was significantly lower in all the ovarian cancer cell lines studied when compared with that in normal ovarian tissues(P 0. 01). ALPL Over-expression resulted in the decreases in cell proliferation and clone formation rate(P 0. 01) and blocked the transition of G1 phase to S phase in SKOV3 cells,as well as suppressed the subcutaneous tumorigenesis in xenografts tumor model(P 0. 05). Conclusion Reversion of ALPL expression level might obviously suppress the cell proliferation and tumorigenesis of SKOV3 cells.
