The study on preparation of superoxide dismutase in erythrocytes

Objective An improved technology for preparation of superoxide dismutase (SOD) from pig or cattle erythrocytes is presented in this paper . Method The designed procedures covered with 60 ℃heat denaturation of autolyzed erythrocytes, removing of hemoglobin with ethanol-chloroform mixture, concentation through rotary vacuum evaporating, precipitation of SOD by (NH4)2SO4 fractinoal salting out and purification of SOD by Sepbadex G-75 gel filtration column chromatography .Result Heat affected the stability of SOD activity and heating autolyzed erythrocytes affected the rate of recovery and purification. It was demonstrated that the method of heat denaturation of autolyzed erythrocytes not only purified SOD but also increased the recovery rate. Conclusion The results indicated that the technological route is rational, practical and of low cost.
