Study of effect of fluoride on the expression of OPG,RANKL and RANK mRNA of rats with fluorosis and the antagonism of Danlan Xianpeng caspule

Objective To observe the expression of 0PG,RANKL and RANK in bone of fluorosis rats caused by water and to explore the antagonism of Danlan Xianpeng Liaofu Jiaonang on the OPG,RANKL,and RANK mRNA.Methods SD rats taken as the experimental objects were randomly assorted into 6 groups(the number either female or male was the same in each group):control group,fluoride group,high-dose medicine group,middle-dose medicine group,low-dose medicine group and borax group.All experimental rats were fed with water which has NaF 50ppm,to establish the animal model of fluorosis.The expressions of 0PG,RANK and RANKL mRNA were observed by RT-PCR.Results Fluoride increased the expression of osteoblastic OPG mRNA and RANKL mRNA and descended RANK mRNA.But OPG mRNA and RANKL mRNA were increased and RANK mRNA was descended in the medicine group.Conclusions Excessive fluoride can induce accelerative bone turnover and enhance the activity of osteoelastic absorption.Danlan Xianpeng Liaofu Jiaonang have antagonism to fluoride and could regulate bone remodeling through 0PG/RANKL/RANK system.
