The fermentive effect on Yersini pestis in sugar and alcohol culture medium of different doses and pH value

Objective Observing and discussing the fermentive effect on Yersini pestis in different culture medium and different doses and PH value so that we can find out the ideal value of Yersini pestis in sugar(alcohol) culture medium.Methods Five kinds of sugar(alcohol) with typical morphological feature of Yersini pestis were selected: Donkey-hide gelatine,rhamnose,maltose,melibiose and glycerol.Use conventional biochemical method to prepare sugar(alcohol) culture medium which is suitable for the growth of plague.According to different doses(0.5 ~1.0g) of sugar(alcohol) and pH(7.2,7.4,7.6),preparing different culture medium and vaccinate plague bacteria,then observe the effect of plague bacteria fermentation.Results Culture medium with 1% sugar(alcohol) and pH value 7.2~7.4 is superior to 0.5% sugar(alcohol) and pH value 7.6,especially for Donkey-hide gelatine and glycerol in fermentive effect on Yersini pestis.Conclusion The ideal value of sugar alcohol medium for Yersini pestis is 1% sugar(alcohol) and pH7.6.
