Influence of low level and long-term arsenic exposure to the phosphorylated protein kinase B as well as its downstream signal factor IKK,I-ΚB and NF-κB in HaCat cells

Objective To discuss the influence of low level and long-term arsenic exposure to the phosphorylated protein kinase B as well as its downstream signaling factor IKK,I-ΚB and NF-κB in HaCat cells. Methods HaCat cells were exposed to NaAsO 2 with different concentration of 0,0. 05,0. 1 μmol / L for 15 weeks,while the protein expression levels of p-Akt,phosphorylated IKK( p-IKK),phosphorylated IκB( p-IκB) and NF-κB were detected by Western Blot. Results Protein levels of pAkt,p-IKK and p-IκB of the 0. 05 and 0. 1 μmol / L groups were significantly higher than that of the control group; protein level of NF-κB in nuclear of the 0. 05 and 0. 1 μmol / L groups were significantly higher than that of control,while its level in cytoplasm were markedly lower than that of control. Conclusion Low level and long term arsenic exposure induces protein phosphorylation of p-Akt in HaCat cells,which result in phosphorylated activation of its downstream signal factor IKK,IκB and expression of NF-κB in the nuclear.
