Effect of lycopene and vitamin E on the expression of clusterin protein in fluorosis rats caused by coal-burning

Objective To study the effect of lycopene and vitamin E on the expression of clusterin protein in fluorosis rats caused by coal-burning.Methods The SD rats were divided into high-,medium-and low-doses of fluorine groups,high dose of fluorine +lycopene group,high dose of fluorine + vitamin E group and normal control group. The rats of control group were treated with normal fodders. The high-,medium-and low-doses of fluorine groups were treated with fodders that with different proportions of fluorine respectively. The rats of high dose of fluorine + lycopene groupwere treated with fodders that with high proportions of fluorine and lycopene. The rats of high dose of fluorine +vitamin E groupwere treated with fodders that with high proportions of fluorine and vitamin E. After 120 d,dental fluorosis was determined by observation,urinary fluoride and testes fluoride were detected by fluorine ion-selective electrode. Hematoxylin and eosin( HE) stain was used to examine the degree of testes injury. The apoptotic index of spermatogenic cell was measured by TUNEL. The expression of clusterin was measuredby immuohistochemistry. Results Compared with the control group,the dental fluorosis,urinary fluoride,testes fluoride,the degree of testes injury and the apoptotic index of spermatogenic cell in high-,medium-and low-doses of fluorine groups were significantly higher( P0.05),and these indicators significantly increased in a dose-dependent manner( P0.05). Compared with the high dose of fluorine group,the dental fluorosis,urinary fluoride,testes fluoride,the degree of testes injury and the apoptotic index of spermatogenic cell in high dose of fluorine +lycopene group and high dose of fluorine + vitamin E group were significantly lower( P0.05). Compared with the control group,the expressions of clusterin in high-,medium-and low-doses of fluorine groups were significantly lower( P0.05),and The expressions of clusterin significantly decreased in a dose-dependent manner( P0.05). Compared with the high dose of fluorine group,the expressions of clusterin in high dose of fluorine +lycopene group and high dose of fluorine + vitamin E group were significantly higher( P 0. 05). Conclusion Coal-burning caused fluorosis may leads to increased apoptotic index of rats' s permatogenic cell,decreased clusterin expression. The antioxidants can decrease apoptotic index of rats' s permatogenic cell,increase clusterin expression,and decrease the degree of testes injury.
