Cloning of Antibacterial Peptide Gene BmCecropinD and Auto-induction Expression of Recombinant Proteins

BmCecropinD is an important antibacterial peptide in silkworm immune system,it has strong antibacterial function.In this study,the total RNA as a template was extracted from the silkworm midgut tissue,then Bombyx mori CecropinD(BmCecropinD)cDNA fragment was cloned by RT-PCR technology.pET32 a-BmCecropinD prokaryotic expression vector was constructed,BmCecropinD recombinant protein was expressed by auto-induction expressing system.The result showed BmCecropinD gene is 245 bp nucleotides in size,and encodes 81 amino acids,the molecular weight of BmCecropinD recombinant protein is 18 ku,the expressing yield of auto-induction is higher than that of IPTG induction.The research laid a basis for purification,activity identification and further application of BmCecropinD antimicrobial peptides.
