Establishment of Identification Method for Infectious Bronchitis Virus Vaccine Strains and Guangxi Field Isolates

According to the conservative sequences of the 3'untranslated region(3'UTR)and the S1 genes of infectious bronchitis virus,two sets of primers were designed to detect the traditional vaccine strains H120,Ma5,4/91,the reference strain M41 and 26 IBV Guangxi isolates by the duplex RT-PCR.The specificity and sensitivity tests were conducted and the duplex RT-PCR was applied to detect experimental samples.The results showed that product of 160 bp was amplified from M41;product of 370 bp were amplified from Mass-type IBV(H120,Ma5,GX-NN2);products of 480 bp and 345 bp were amplified from 4/91-type IBV(4/91,GX-NN7);products of 260 bp to 345 bp were amplified from the other 24 Guangxi IBVs.NDV,ILTV,MDV,IBDV and ALV were not amplified by the duplex RT-PCR.The lowest detected template is 100 pg.90% experimental samples were positive detected by the duplex RT-PCR.The present study showed that the established differentiation method for distinguishing Guangxi field strains from commercial vaccine strains of IBV has characteristics of strong specificity and high sensitivity.
