Study on Anti-inflammatory Effect of YuxingDangshen-San in Vitro

To explore the anti-inflammatory effect of natural forage plant formula YuxingDangshen-San in vitro,this experiment evaluated the anti-inflammatory effects of YuxingDangshen-San by observing the effect of it on NO,TNF-α,IL-1β,IL-6 and IL-10 levels in culture supernatant of RAW264.7 cells activated by lipopolysaccharides(LPS),and the protein expression levels of P65 and IκB in nuclear factorκB(NF-κB)signal pathway.The model of inflammation was made by RAW264.7 cells activated by lipopolysaccharides.Cells at logarithmic growth phase were allocated to blank control group,model group(treated with LPS),YuxingDangshen-San group(treated with 5 mg/mL,2.5 mg/mL,1.25 mg/mL YuxingDangshen-San respectively).After being treated with YuxingDangshen-San for 4 hand then incubated with 1μg/mL LPS for 24 h,the change of NO level was observed by Griess assay,the levels of TNF-α,IL-1β,IL-6 and IL-10 were evaluated by ELISA,the levels of P65,IκB,P-P65 and P-IκB were evaluated by Western-blot.The experimental data showed that after RAW264.7 cells were activated by lipopolysaccharides,YuxingDangshenSan can inhibit the increase of TNF-α,IL-1β,IL-6 levels significantly(P 0.01)with a dose-dependent manner in certain degree.It can reduce the phosphorylation levels of P65 and IκB obviously(P0.01).The results showed that YuxingDangshen-San has good anti-inflammatory effects in vitro.
