Comparison of ITS1 Intra-Individual Variability of Ascaris suum

ITS1 sequences are often used for genotyping,species identification and phylogenetic research.This paper compares the individual ITS1 sequence of Ascarissuum.DNA was extracted fromA.suum(n=5)collected from Jiangxi Province.The ITS1 fragments of rDNA were amplified by PCR and the PCR products were cloned and sequenced.MEGA5.0 was used to sequence analyses.The results indicated that the copies of ITS1 sequence of the same sample are not entirely consistent.There is variability,particularly between the polyA and polyT.One sample contains multiple Ascaris genotypes or haplotypes.New haplotypes(HC1-HC5)were discovered in the experiment.Therefore,using this genetic region to discriminate genotypes of A.suum should be reconsidered.
