DRM1,DRM2 are involved in the callus formation in Arabidopsis

By stereo microscope,scanning electron microscope and transmission electron microscope,the microstructure and ultra structure of cells from callus can be observed. Results show that the seedlings of Arabidopsis mutant drm1 / drm2,lacking the de novo DNA methyltransferase DRM1,DRM2,can be induced to form more callus compared to the wild type( WS) on Callus-inducingMedium( CIM,containing 1 mg / L 2,4-D,0. 1 mg / L KT). Other different explants of roots,leaves and petioles were used to compare with the seedlings and obtained the same results. The molecular results show the expression levels of genes DRM1,DRM2coding the de novo DNA methyltransferase DRM1,DRM2 are related to the concentration of hormones( auxin and cytokinin) which can induce the formation of callus. So it can be indicated that DRM1,DRM2 are possibly involved in the callus formation.
