Influence of LaCl_3 on the morphology of pollen tube and distribution of wall components of Qinguan apple

In this study,the effect of LaCl3,calcium channel blocker,on apple pollen tube and wall components was investigated.Results illustrated that LaCl3 inhibited the pollen germination and tube growth,resulting in abnormal pollen tube. The cytological results indicated that the pollen tube wall was mainly composed of cellulose,callose,pectins and arabinogalactan proteins( AGPs).Acidic pectin and esterified pectin were uniformly distributed on the control tube wall,but on the LaCl3-treated tube wall,acidic pectin and esterified pectin accumulated in the apex. AGPs were distributed mainly on the basal part of the tube in control pollen,but LaCl3-treatment made AGPs accumulated on the whole tube. FTIR microspectroscopy illustrated that LaCl3 induced obvious changes in the composition of pollen tube walls. There was an increase in acid pectin and proteins,which was in agreement with the result of immunolabeling with JIM5 and LM2. In a word,the results suggested that laCl3 induced the changes in the composition of the tube wall,which further ceased the growth of pollen tube.
