Detection of tomato zonate spot virus by immunogold labeling electron microscopy

In this study,the immunogold labeling electron microscopy was used to detect Tomato zonate spot virus( TZSV). The polyclonal antibody against TZSV N protein and the polyclonal antibody against TZSV Gn protein were used as primary antibody,respectively,and the colloidal gold labeleld Goat anti-rabbit Ig G-gold( 10 nm) was used as the secondary antibody. Under the electron microscopy,TZSV particles labelled with colloidal gold particles could be observed when the polyclonal antibody against TZSV Gn protein was used as primary antibody. The result indicated that it was an accurate and reliable method to detect TZSV.
