Using mixed cellulose ester membrane to prepare liquid algae samples for cryo-scanning electron microscopy

To optimize the sample preparation conditions of Cryo-scanning electron microscopy( Cryo-SEM),we compared the enrichment and Cryo-SEM observation effects of mixed cellulose ester membrane( MCEM) and filter paper for liquid algae cells.Microcystis aeruginosa and Chlorella( diameters about 2 to 3 μm) were chosen as materials. The pores of MCEM were small and uniform in distribution. Both front and back surfaces of MCEM could effectively adsorb algae cells. MCEM could adsorb more cells and get better imaging effects than the filter paper. Cryo-SEM images showed that cells on MCEM were plump in morphology,and with no distortion or shrinkage. MCEM was more suitable for micro liquid algae samples preparation for Cryo-SEM.
