Proteome-Wide Identification of poly(ADP-ribose)Binding Proteins

ADP-ribosylation is a post-transcriptional modication and plays important roles in a variety of cellular processes.Many PARylated proteins and several PAR binding modules have been identified recently.Here we describe a sensitive proteomics approach based on mass spectrometry to identify PAR binding proteins.While confirming many of the known PAR binding proteins,we identified numerous additional PAR binding targets.In vitro validation of four PAR binding candidates confirmed that our methodology targeted PAR binding proteins.We also found that HRP-2 interacted with PAR via its PWWP domain,and further demonstrated that the PWWP domain was a PAR binding module.Moreover,HRP-2 depletion cells were sensitive to DNA damage agents,suggesting that HRP-2 participated in DNA damage response.
