Isolation,Identification and Heavy Metal Tolerance of an Acidophilic Heterotrophic Microorganism

An acidophilic heterotrophic microorganism JJU-2 was isolated from sewage sludge which was collected from the sewage disposal plant.According to its morphology,physiological-biochemical properties and 26 S rDNA gene sequence analysis,the strain was identified as Rhodotorulamucilaginosa.The optimal pH of strain was 5 but it could survive in an extremely low pH 1 condition.The optimum growth temperature was 25 t and the highest temperature it can survive was 40 t.The strain grew fast after 4 h and came into the logarithmic growth stage until the stable growth stage after 16 h.Resistant experiment to metals showed that JJU-2 strain could resist cadmium,chromium,copper,nickel and lead with 1.5 mmol/L,10mmol/L,5 mmol/L,2 mmol/L and 20 mmol/L,respectively.The leaching rates of heavy metals(Cu,Ni,Cd and Cr) were84%,96%,100%and 71%with the combined effect of JJU-2 and JJU-1 strains,respectively.The isolation of the strain Rhodotorulamucilaginosa could be widely used in sewage sludge bioleaching of heavy metal and provides basic material to study the resistance mechanism of microorganisms to heavy metal in extreme conditions.
