Effects of 1,3-Dibromo-5,5-Dimethylhydantoin on Gene Expression of Antioxidant Enzyme in GIFT Tilapia Plasma

The purpose of this study was to find the steady biomarkers by studying plasma gene expression profiles in male GIFT tilapia(Oreochromis niloticus),which has been treated with different concentrations(0,0.06,0.3,1.5 mg/L 1,3-dibromo-5,5-dimethylhydantoin)DBDMH exposure at different time points(1,2,4,8,16 days and with the repair period on 26 days). The study on gene expression profile was performed using real-time PCR method,and the detected genes were as superoxide dismutase(sod), glutathione-S-transfer(gst),glutathione reductase(gr),glutathione peroxidase 1(gpx1)and catalase(cat). The plasma antioxidant enzyme transcripts of male GIFT tilapia were significantly induced under 0.3 mg/L DBDMH exposure except for gr. To different DBDMH concentration groups,plasma transcripts at 1 d(sod,gst,gpx1)and 2d(gst,gr),4 d(sod,gpx1) showed dose-dependent decreases and increments,respectively. Gene expression of such these genes(gst,gr,gpx1) in 0.3 mg/L DBDMH exposure groups were significantly lower than those in 0.06 mg/L DBDMH exposure groups. All values of those genes were not back to the normal level in 0.3,1.5 mg/L DBDMH concentration groups after the recovery period for ten days. These results suggest plasma antioxidant enzyme transcripts(except for gpx1,cat at1 d;gr at 8 and 16 d)of male GIFT tilapia are significantly increased under 0.3 mg/L DBDMH exposure,and those detected genes could be used as the sensitive biomarkers.
