Optimization of Astaxanthin Extraction Technology from Procambarus clarkii

The astaxanthin was extracted from fresh Procambarus clarkii shell, the effect of extraction solvent, solid-liquid ratio, extraction time, extraction temperature and extraction times as the factors on the extraction efficiency were studied.Freeze-dried, room temperature dried and oven dried as the sample pretreatments on the extraction efficiency of astaxanthin were compared by single factor experiment. The astaxanthin content and standard recovery rates were measured by UV-Vis spectrophotometry. The results showed that, acetone was more effective than ethanol or petroleum ether on the extraction of astaxanthin. Freeze-dried sample pretreatment method was the best choice. The optimal conditions of the astaxanthin extraction were extracting for 2 h each time at 30 ℃ with solid-liquid ratio of 1∶20(g∶mL), extracting twice. The results of standard recovery rates on optimal extraction conditions was 99.2% and the mean contents of astaxanthin extracted from the fresh Procambarus clarkii shell was 148.2 μg/g.
