Research on open-ended photoautotrophic micropropagation technology of Acacia melanoxylon

In order to simplify the procedure of photoautotrophic micropropagation,the research of Acacia melanoxylon's photoautotrophic micropropagation was carried out in the open container,by adding 5 mL tuzet (500 mg·kg-1) to agar medium.The results showed that by means of open-ended photoautotrophic micropropagation,after 21 days,A.melanoxylon's rooting rate amounted 85%,while its contamination rate was 9.2%;and that its chlorophyll content and aboveground biomass were higher than those of its counterparts under the conditions of sand-culture-medium photoautotrophic micropropagation and under traditional culture;and that its underground biomass and transplanting survival rate were correspondingly lower than those under the condition of sand-culture-medium photoautotrophic micropropagation,but higher than those under the condition of the traditional culture.
