Extraction and Analysis of Genomic DNA from Leaves of Castanopsis carlesii

In this study,taking the leaves of carlesii community of Castanopsis carlesii as materials,the modified CTAB method was adopted to extract the genome DNA of Castanopsis carlesii,and the obtained genomic DNA was tested for concentration,PCR amplification,agarose gel electrophoresis detection,content determination analysis and amplification result sequencing.The results showed that the quality and concentration of genomic DNA extracted from Castanopsis carlesii leaves were good,with good integrity,good PCR effect,clear display of electrophoretic mapping strips,and the length of sequencing results met the requirements.The DNA extracted by this method can be used for the analysis of genetic diversity of Castanopsis carlesii,which will provide reference for the DNA extraction and other molecular research of other Castanopsis plants,such as Castanopsis carlesii and Castanopsis kawakamii.
