Morphology of Pollen Grains from the Plants with Different Type of Sex by Scanning Electron Microscope(SEM) and the Viability of Pollens in Red Bayberry

Ten pairs of polymorphic SSR primers were used for fingerprint of three types of male flower from the plants with different sex in Cixi red bayberry,including androphyte,female individual with a mutant branch of staminate inflorescence and monoecious plant.The results showed that the female individual with a mutant branch of staminate inflorescence was the variety of Biqi,a popular cultivar.By scanning electron microscope,we found the pollen size were in the range of(22.9~23.6)×(20.3~22.1) μm,and the diameter of the three typical germinal poles were 2.90~3.03 μm without significant difference.Monoecious plant showed the largest pollen viability tested by both germination and I-KI staining.The most suitable culture media for pollen germination was 5% sucrose + 0.01% boric acid + 1% agar.
