This study is to investigate the changes of GADD45 and CDKN1A gene mRNA expression induced by 60Co γ-ray in lymphoblastoid cell line in vitro,and its probability of gene expression as a biodosimeter.Lymphoblastoid cell line was irradiated by 60Co γ-ray with the dose of 0-10 Gy and cultivated for 8 hours,in order to screen the optimal dose.mRNA was extracted and real-time PCR method was used to detect the changes of PIG3 and GADD45 gene mRNA expression in lymphoblastoid cells.Lymphoblastoid cells was cultivated for 0-72 h after optimal dose irradiation.The changes of GADD45 and CDKN1A gene mRNA expression in lymphoblastoid cells in different time-point were detected.The results showed that GADD45 and CDKN1A gene expression in lymphoblastoid cells increased in a dose-dependent form.The peak expression of GADD45 was at 8 h after 5 Gy irradiation and began to decrease.The trend in change of CDKN1A gene expression was not notable.
