Experimental study of effect of ligustrazine on the thrombosis of PTRA site in canine renal artery stenosis model

Objective:To evaluate the effect of ligustrazine on preventing the thrombosis at the site of percutaneous transluminal renal angioplasty (PTRA).Methods:Dogs models (n=15) with experimental renal stenosis were selected and randomly divided into control group,ligustrazine group and aspirin group.Each group had five dogs.PTRA were performed 6~8 weeks after experimental renal stenosis.All dogs were killed 4 hours after PTRA.Angioplastic renal arteries were removed before dogs were killed.Renal specinens were fixed for light and electron microscopic analysis.The thrombus volume and microstructure of platelet thrombus at PTRA sites were measured and analysed respectively.Results:The thrombus volume of the two experimental groups were significantly smaller than that of the control group.Compared with the two experimental groups,the fibrin area were significantly high,the gap area were significantly low in the white thrombus in the control group;the number of platelet and thread granules were evidently high,but relative number of α granules and compact granules were significantly low in the control group.Compared with each other,the changes of the thrombus volume of PTRA sites,the fibrin and gap area in the white thrombus,the number of platelets and thread granules,relative number of α granules and compact granules were not statistically significant in two experimental groups.Conclusion:Ligustrazine could be one of the effective drugs in preventing thrombosis after PTA.
