Dynamic Study on Cytosine Deamination by Capillary Electrophoresis

Cytosine deamination has been considered to be one of the key reasons for DNA damage. In the paper, we first developed a sensitive and highly selective method to determine contents of cytosine and uracil based on capillary electrophoresis with a UV absorbance detector. Then, we used this method to investigate the cytosine deamination to uracil in the presence of hydrogen peroxide. Data obtained showed that the reaction orders for cytosine and hydrogen peroxide were both one order and the activation energy was 70.35 kJ·mol -1, lower apparently than that of spontaneous reaction.
