Determination of Trace Pyrophosphate by Bioluminescence

A new method for the analysis of inorganic pyrophosphate (PPi) by utilization of ATP sulfurylase and luciferase is described. In the assay, PPi in the samples is converted to ATP by ATP sulfurylase, then ATP can be detected by ATP bioiuminescent assay kit. The PPi contained in the reagent can be removed by a long incubation with a low concentration of PPase. The assay is extremely sensitive and rapid. The linear ranges is 1×10 -9 ~ 5×10 -7 mol/L, and the detection limit is 2.8×10 -11 mol/L.
