Aptamer Functionalized Nanocomposites Targeted Thrombin for Anticoagulation in the Cardiovascular Diseases Therapy

Novel aptamer-functionalized magnetic beads/gold nanorods nanocomposites were created for reversible regulation of thrombin activity.By utilizing proximity-dependent surface hybridization method,a closed-loop structure can be constructed with thrombin and its couple aptamers.With this high stable structure,the binding effect between aptamers and thrombin can be greatly enhanced.Considering the excellent localized surface plasmon resonances(LSPRs)property of gold nanorods(Au NRs),the activity of inhibited thrombin is efficiently recovered under near-infrared light stimulation by using Au NRs as photothermal agents to disrupt the closed-loop structure.Real-time light scattering spectroscopy and scanning electron microscopy have been used to determine thrombin activity.Thrombin clotting time performed in human plasma further demonstrated that the regulation method held great promise for potential clinical application.Furthermore,the artificial dialysis simulation experiments showd this method could also be applied to potential areas such as the hemofiltration of renal failure therapy.
