Construction of a Genomic DNA Library of an Indica Rice Variety H359 Based on Transformation-Competent Artificial Chromosomes

Construction of artificial chromosome carrying large inserted DNA fragments that can be transferred into plant genomes mediated by Agrobacterium could speed up the work of fine mapping and positional cloning of mutant genes. To facilitate the cloning of mutant genes, a genomic DNA library was constructed for an indica rice variety H359 using vector pYLTAC7. The library consists of 41088 individual colonies stored as frozen cultures in 107 plates with 384 wells each. The sizes of inserts are 50 100kb and the average is 77kb. The rice genome coverage of the library was estimated to be about 7 4 haploid genome equivalents.
