Cloning and Sequence Analysis of Transcription Factor Gene Ta TGA4 from Wheat-Thinopyrum Alien Addition Line

In this study, a novel gene fragment that is up-regulated in response to powdery mildew bacterium E09 was identified through the Genefishing technology. And the full gene, encoding transcription factor Ta TGA4, was cloned from germplasm SN6306, a hybrid Triticum aestivum-Thinopyrum intermedium alien disomic addition line with highly resistant to powdery mildew. Sequence analysis of the c DNA and the predicted protein shows that the ORF of the Ta TGA4 gene is 1 221 bp in length and encodes a protein with 406 amino acid residues. BLASTP analysis shows that Ta TGA4, containing two domains, has 92% identities with the homologous protein from Aegilops tauschii as well as 90% identities with the homologous protein from Triticum urartu. Based on the results of multiple alignments and phylogenetic analyses, it was deduced that Ta TGA4 should belong to TGA4 transcription factor. This research would lay the foundation for further functional analysis of Ta TGA4 in powdery mildew resistance.
