Application of Gene-deletor System to Create Multiple Stress Tolerant Corn New Germplasm with Gene ZmSDD1,BT and BAR

We transformed plant expression vector that containing pollen specific promoter Zm13 driven Gene-deletor exogenous gene deletor system, maize Ubi promoter-driven gene expression Zm SDD1, Actin promoter-driven Bt gene and Ubi driven Bar::GUS fusion gene to maize Jiao51 by Agrobacterium-mediated transformation of maize shoot tip in this study. By screening, GUS staining and PCR analysis, we identified 31 transgenic plants. We found drought tolerance of transgenic plants improved than wild-type plants, and improved transgenic maize insect- and herbicide-resistance. Drought resistance identification showed that survival rate of transgenic plants was 70% higher than wild type after drought treatment, at the same time, transgenic maize leaf water retention was significantly higher than wild-type plants. Insect resistance test indicated that transgenic plants has obviously resisted to corn borer, also inhibited feeding and development of corn borer. Leaf in vitro and field herbicide resistance test suggested that transgenic plants had a higher tolerance concentration than wild type,maximum resistance to 150 mg/L. Determination of transgenes excision efficiencies in 20 strains of transgenic maize plants, it found that some plants can completely eliminate exogenous gene from maize, while that not removed completely need to further research. This study created a drought-, insect- and herbicide-resistant complex new germplasm that lay the foundation for cultivating multiple stress tolerance corn and proved that exogenous gene could be cleared compeletey by "Gene-deletor" technology in transgenic corn, which provided technical support for transgene-safty.
