Genetic Structure of Five Dioecious Industrial Hemp Varieties in Yunnan

To reveal genetic structure of dioecious industrial hemp, population diversity and genetic differentiation of five typical dioecious hemp varieties in Yunnan were studied using RAPD and ISSR molecular markers. Four RAPD primers and eight ISSR primers amplified 74 loci with 64 polymorphic bands in 120 individuals from five industrial hemp populations. The polymorphic rate was 86.5%. The Nei's gene coefficient of the total gene diversity(He) was 0.309 5, and the Nei's gene diversity within populations ranged from 0.216 7 to 0.287 8 with the average value of 0.251 3. The level of genetic diversity can be ranked as: Yunma No.3 Yunma No.4Yunma No.1 Yunma No.5 Yunma No.2. The genetic similarity coefficient among populations ranged from 0.874 9 to0.925 5, and the gene flow among populations(Nm) was 2.159 5, which meant that the five industrial hemp varieties showed higher genetic homogenization. The coefficient of genetic differentiation(Gst) among the five populations was 0.188, which meant gene diversity among populations accounted for 18.8% of all the diversity of populations.This value is higher than that of outcrossing and wind pollination crops(average Gst=0.100). It can be inferred that artificial selection had widened the genetic differentiation in the breeding of industrial hemp. The results show that the genetic similarity in dioecious hemp variety population is higher, and it has great potential to develop the breeding of hemp through purifying selection within population or using special resources with large genetic distance.
