Physical Localization of SBE and GBSS Genes from Cassava(Manihot esculenta Crantz)

Starch branching enzyme(SBE) and Granule bound starch synthase(GBSS) genes are key enzymes of starch synthesis processes in cassava.In this study,Fluorescence in situ hybridization(FISH) was used to locate the SBE,GBSS Ⅰ and GBSS Ⅱ genes which related to starch synthesis of cassava.The results indicated that the GBSS Ⅰ and GBSS Ⅱ genes were located on the short arms of chromosome 13 and 11 in Cassava M.SC6.The percentage distances from centromere to signal were 26.3 ±0.1 and 14.0 ±0.5,and the signal detection rates were12.9% and 7.9%,respectively.The SBEI gene was located on the long arm of the chromosome 12 in M.SC6,the percentage distance from the centromere to signal was 76.2 ±0.2,and the signal detection rate was 9.4% as well.The linkage analysis was proceeded with the located genes,then we found SBEI,GBSS Ⅰ and GBSS Ⅱ genes were separate genes since they distributed on different chromosomes of Cassava.The physical location of those genes will reveal the linkage relationship and the characteristics of distribution between them and other function genes,which effectively apply to molecular assisted breeding and comparative genomics of the Cassava.
