Isolation and Characterization of Cu/Zn Superoxide Dismutase Gene in Saccharum arundinaceum Retz

Saccharum arundinaceum Retz.is one of the most important wild sugarcane germplasm having strong resistance,which is great significance for sugarcane breeding by researching and using its resistant gene.In this research,two complete coding sequences(SaSOD-1a and SaSOD-1b,Gen Bank Accession number KJ001795 and KJ001796) of Cu/Zn Superoxide Dismutase gene were cloned from Saccharum arundinaceum Retz.by blasting the sugarcane EST database using sorghum Cu/Zn-SOD gene(Gen Bank Accession number XM 002445626.1) cDNA sequence as a querying probe.SaSOD-1a gene full-length sequences were 2 473 bp,and SaSOD-1b gene's 2 228 bp.Two full-length sequences contain 7 introns and encode 206 amino acid.Between these two sequences was 98.6%similarity,there are 8 single nucleotide mutation(SNP) sites,protein 99.0% similarity,there are 2 amino acid mutation sites.The putative SaSOD-1a and SaSOD-1b protein sequence and other species homologou s evolutionary analysis,have shown a high conservatism.The study laid the foundation for the further understanding of the relationship between superoxide dismutase and Saccharum arundinaceum Retz.drought tolerance.
