Cloning and Characterization Analysis of a New Endoglucanase Gene OsGLU16 in Rice(Oryza sativa L.)

β-1, 4-endoglucanase(endo-β-1, 4-glucanase, EGase) plays an important role in plant tissue growth,flowering, fruit ripening, organ shedding and so on. In this study, we cloned a new gene called Os GLU16 fromβ-1, 4-glucan endonuclease family in rice. Sequence analysis showed that this gene was located on chromosome8, and the full length of c DNA was 1 827 bp. It contained an open reading frame of 1 572 bp, encoding 523 amino acid residues. Protein homology analysis indicated that the protein encoded by the Os GLU16 gene had a highly conserved GH9 catalytic domain in the 54~516 amino acid residue region. We successfully constructed Os GLU16 overexpression vector p Ca MV35 S-Os GLU16-EGFP and obtained transgenic rice plants. The phenotypic analysis suggested that the overexpression lines had a significant decrease in plant height and spike length compared withwildtype. This study has preliminarily elucidated the phenotypic characteristics of Os GLU16 gene overexpressing transgenic rice plants, which might provide a theoretical basis for further enriching the role of β-1,4-glucan endonuclease gene family in rice growth and development.
