Expression of the Transcription Factor LbMYB103 of Lycium barbarum in Transgenic Tobacco

To investigate the effect of the tran scription factor Lb MYB103 of Lycium barbarum during the development of plant, we isolated the complete open reading frame(ORF) from the anthers of Ningxia L. barbarum'Ningqi No. Ⅰ' by RT-PCR technology, and constructed plant over-expression vector p Cambia2301-ky-Lb MYB103 which was transformed into tobacco mediated by Agrobacterium. T0 transgenic positive plants were obtained by resistance screening and PCR identification. After resistance screening and PCR identification, we got genetically stable T1 and T2 positive transgenic tobacco from T0 positive tobacco seeds. We observed the plant phenotypic traits during the growth and development. The results showed that the exogenous Lb MYB103 was successfully integrated into transgenic plant genome and could be expressed in strong promoter Ca MV35 S.Compared with the wildtype, the transgenic tobacco grew slowly, and the plant height was about half of the wildtype. The floral organ development was abnormal, but part of the petals were aberrant. The results indicate transcription factor Lb MYB103 might play a regulatory role in the development of tobacco floral organ, which could provide a preliminary theoretical basis for further study of Lb MYB103 in the regulation of the floral organ development of L. barbarum.
