Construction of Genetic Map for Rice Doubled Haploid Population by Specific-length Amplified Fragment Sequencing

A doubled haploid(DH) population constructed by an indica cultivar ZYQ8 and a japonica cultivar JX17 was sequenced by specific-length amplified fragment Sequencing technology. A total of 70 997 SLAF markers were obtained by comparing the sequenced data with rice reference genome. The genetic map for the DH population was constructed by 8 940 selected high quality SLAF markers. The genetic map covered 12 linkage groups and was 2 294 71 c M in length with an average distance of 0.26 c M between adjacent markers. The statistic analysis between the linkage groups of the genetic map and the rice physical map showed that the correlation coefficient of the chromosome 9 was 0.87, and correlation coefficients of the other eleven chromosomes were all above 0.95, indicating a great collinearity relationship between the 12 linkage groups of the genetic map and the rice physical map. The high density genetic map might provide a platform for further study of specific genes in indica and japonica cultivars.
