Cloning and Bioinformatics Analysis of RcGA20ox1 Gene from Castor

GA20 oxidas e is an important oxidase in the gibberellin biosynthesis pathway. We try to clone this enzyme to discover the candidate gene of GA20ox family in castor, and to speculate its function. According to the known castor gene sequence of GA20 ox1(XM_002510827.2) on NCBI, primers were designed and amplified to get GA20ox gene fragments. Connected with p MD18-T vector, the study obtained the complete open reading frame(ORF) sequence in 1 137 bp of RcGA20ox1 cDNA. The sequence was compared, while the physical and chemical properties of transcripts, hydrophobicity, functional domain were analyzed and their tertiary structure was predicted. It was presumed that it could encode 378 amino acids and was a hydrophilic stable protein. The gene belonged to the Pcb C superfamily, which was a non-secreted protein and cytoplasmic protein and contained 28 phosphorylation sites. The predicted structure was composed of 8 α-helices and 8 β-folding, and was highly homologous with the Brazilian rubber tree, cassava and Jatropha curcas. This study might provide a theoretical and practical basis for the castor GA biosynthesis and even castor dumbing.
