Cloning and Expression Vector Construction of miR164 and Its Target Gene NAC in Wheat

miRNA plays an important role in plant growth regulation and adversity adaptation. In this study, in order to explore the function of miR164 gene and its response mechanism toward stress, we firstly monitored the expression change of miR164 and its target gene NAC2D in wheat under endoplasmic reticulum(ER) stress, and then expression vectors were constructed by cloning miR164 and several genes of NAC family(NAC6A, NAC2D,NAC8 and NAC1), respectively. The results showed that the expression of miR164 was up-regulated and NAC2D was down-regulated after being treated with dithiothreitol(DTT) which was an inducer of ER stress, but the expression pattern of both genes exhibited a reverse trend after being treated with ursodeoxycholic acid(UDCA)which was a reliever to assuage ER stress, indicating that miR164 might be involved in the regulation of ER stress response in wheat. Moreover, several genes of NAC family were cloned respectively, and their expression vectors were successfully constructed, which would provide an important foundation for further exploration of the function of miR164 and its regulatory mechanism.
