Callus Induction and Multiplication Culture of Herbaceous Stem of Ephedra sinica Stapf

One or two year old herbaceous stems of Ephedra sinica were selected as explants, and different combinations of sterilization and different concentrations of hormones were used to investigate the optimum explant culture method and the optimum medium combination for callus induction and proliferation. The results showed that the best combination of explant sterilization was 75% ethanol sterilization for 20 s and then 0.1%HgCl_2 for 4 min, and survival rate of this sterilization combination was up to 100%, with no deaths and contamination. The best callus induction medium of herbaceous stem of Ephedra sinica was MS+6-BA 0.25 mg/L+NAA0.05 mg/L; the best callus proliferation medium was MS+6-BA 2.0 mg/L+NAA 0.5 mg/L; the inducing effect of annual herbaceous stem was significantly better than that of the biennial stem. This study could provide theoretical and technical basis for industrialized seedling production and secondary metabolite production of Ephedra sinica.
