Optimization of Polyphenols from Hydrocotyle sibthorpioides by UltrasonicAssisted Extraction Using Response Surface Methodology and Its Antioxidant Activities

This study aimed to optimize the extraction technology of polyphenols from Hydrocotyle sibthorpioides.Hydrocotyle sibthorpioides was selected as the raw material, the polyphenols yield rate was set as the response value, and four factors(ethanol concentration, ultrasonic time, liquid-to-material ratio and ultrasonic temperature)which affect the polyphenols yield rate of Hydrocotyle sibthorpioides were investigated. Also, the extraction processes were designed and optimized by Box-Behnken experiment, so as to evaluate its antioxidant activities by the clearance rate of hydroxyl radicals. The results showed that the optimal extraction conditions of polyphenols from Hydrocotyle sibthorpioides were ethanol concentration 64%, ultrasonic time 34 min, liquid-to-material ratio23 m L/g and ultrasonic temperature 76℃. The yield of polyphenols could be up to 37.94 mg/g under the optimal condition, and the relative error was 0.52% compared to the predictive value(38.14 mg/g) which indicated the feasible model fitted well with the experimental data. The polyphenols from Hydrocotyle sibthorpioides had certain antioxidant ability and its antioxidant capacity had a positive correlation with the concentration. The technology provided the scientific basis for the extraction and utilization of polyphenol from Hydrocotyle sibthorpioides.
