THE INVESTIGATION OF HEPATOCYTE APOPTOSIS AND ITS MODULAT- ING FACTORS IN CIRRHOTIC LIVER TISSUES

Objective To investigate the death patterns of hepatocyte in patients with cirrhosis. Methods 20 liver specimens of cirrhostic patients were collected during operation and other 5 controls from patients with liver he-mangioma. Hepatocyte apoptosis were identified under light microscopy,electric microscopy and measured by flow cy-tometry.The affecting factors of apoptosis such as Bcl-2,C-myc,C-fos,TGFβ,TNFα、iNOS were also estimated by immunohistochemistry. Results There were two different death patterns simultaneously in these cirrhotic tissues: apoptosis and necrosis. The amount of apoptosis remarkably increased in cirrhosis group with comparison to that of controls with the apoptosis index of 3.06 +/-0.79%to 0.56+/-0.20%,respectively( P 0.05) .The expressions of Bcl-2,C-myc,C-fos,TGFβ,TNFα,iNOS were positive or densely positive in cirrhotic group,while negative in normal liver tissues. Conclusion Apoptosis is one kind of death patterns in liver cirrhosis due to viral hepatitis and plays an important role in the pathogenesis of cirrhosis. The apoptosis may be related to certain genes and cytokines such as Bcl-2,C-myc,C-fos and TGFβ,TNFα、 iNOS.
