Effects of Cumulus Cells on in Vitro Maturation,Fertilization and Embryo Development of Procine Oocytes

In this study, porcine oocytes collected from overies were divided into two groups according to the thickness of their cumulus cell layer. Group 1 of oocytes have 3 or more than 3 layers of cumulus cells, Group 2 of oocytes have 1-2 layers. These two groups of oocytes were used for maturation culture, in vitro fertilization(IVF) and partheno-genetical activation(PA) tests to compare their developmental potential. The results showed that oocytes of Group 1 had significantly higher(P0.01) maturation rate than that of Group 2(67.42±1.52%vs 48.33±2.85%), and PA embryos generated from Group 1 oocytes had significantly higher(P0.01) blastocyst rate than that of PA embryos produced from Group 2(58.00±2.88% vs 41.00±6.40%) although the blastocyst total cell number was similar(P 0.05) between the two groups The blastocyst rate(18.00±5.70% vs 4.25±2.31%) and the blastocyst total cell number(41.7±3.33 vs 36.2±2.10) of IVF embryos generated from Group 1 oocytes were significantly higher(P0.01) than that of Group 2. Moreover, IVF embryos generated from cumulus cells-enclosed oocytes have a higher(P0.01) cleavage rate(88.14±7.48% vs 58.69±2.89%), a higher(P0.01) blastocyst rate(51.2±7.33% vs 11.92±2.29%) and higher(P0. 01) blastocyst total cell number(41.7±3.33 vs 36.8±3.15), compared to IVF embryos produced from cumulus-denuded oocytes. These results suggest that oocytes having thicker layer of cumulus cells bear higher maturation rate and higher embryo developmental ability after IVF and PA, and the presence of cumulus cells during oocyte fertilization was helpful to promote the developmental potential of IVF embryos.
