Preventive Effect of Fonzylane on Nuclear Translocation of Apoptosis-induced Factor in Neuron-like Cells Differentiated from PC-12 Cells

Objective: To investigate the molecular mechanisms that Fonzylane inhibits apoptosis of neuron-like cells differentiated from PC12 cells induced by hypoxia.Methods: Neuron-like cells differentiated from PC12 cell model were exposed to hypoxia for 24 h. The serum containing Fonzylane was made to pre-protect the cells before exposure to hypoxia.When compared with the un-treated cells,apoptosis-inducing factor(AIF) localization was explored by immunofluorescent staining.In addition, the expression of heat shock protein 60(HSP60) was studied.Results: AIF was released from mitochondria and translocated to the nuclei of the control cells.Whereas the cells pretreated by the Fonzylane-containing serum for 24 h did not show any AIF translocation to the nuclei.Conclusion:By blocking the AIF translocation to the nuclei following hypoxia effectively,Fonzylane could suppress the apoptosis of neuron-like cells differentiated from PC12 cell.
