A Study on Mechanism of Pizotifen and Propranolol for Prophylaxis of Migraine

Objective:To explore mechanism of prophylactic effect of pizotifen and propranolol on migraine.Methods:Fifty SD rats were randomly divided into the groups control,normal saline(NS),model,pizotifen and propranolol.A Migraine model was generated.The groups pizotifen and propranolol were fed with pizotifen or propranolol respectively after the second operation.After the fifth operation,the content of CGRP in the jugular vein and the expression of α-CGRP mRNA in the brainstem and trigeminal ganglion have been determined,and the behavior of the rats at the same time was studied.Results:The tolerance for nitroglycerin of the rats in groups pizotifen and propranolol were enhanced when compared with model group.There was no significant difference in the content of CGRP among the groups(P0.05).The expression level of α-CGRP mRNA in the brain stem and trigeminal ganglion in groups pizotifen and propranolol was decreased significantly when compared with other groups(P0.05).Conclusion:The prophylactic effect of pizotifen and propanolol for migraine may relate to inhibition of α-CGRP mRNA expression in the brainstem and trigeminal ganglion.
