Protection of Fasudil on Spinal Ischemia-reperfusion Injury in Rabbits

Objective:To explore the neuroprotective effect of Fasudil,a Rho-kinase inhibitor,on rabbit spinal ischemia-reperfusion injury.Methods:The ischemia-reperfusion model of the spinal cord was conducted by the selective occlusion of the lumbar arteries in thirty rabbits,which were randomly divided into Fasudil(n=20) and control groups(n=10).The rabbits in the Fasudil group were given Fasudil through intraperitoneal injection whereas the control group received the equivalent physiological saline in the same way.After two weeks,the level of free ra-dical(MDA,SOD and NO) in the serum was detected,and the function of hindlimbs was evaluated by the Jacobs method.In addition,the motor evoked potentials(MEP) were also assessed.Results:Treatment with Fasudil resulted in a significant improvement of Jacobs score(P0.001),demonstrated by the decreased levels of free radicals when compared to that in the control group(P0.05 or 0.01).The MEP in the Fasudil group was shorter than that in the control group(P0.001).Conclusion:Fasudil provides a neuroprotective effect on rabbits following spinal ischemia-reperfusion injury.
