White Matter Damage in Mice with Bilateral Common Carotid Artery Stenosis

Objective:To establish a mouse model of chronic cerebral hypoperfusion white matter damage using bilateral common carotid artery stenosis(BCAS). Methods: Forty-two C57BL/6J mice were randomly divided into sham group(n=21) and BCAS group(n=21). The mice in BCAS group were used to establish chronic cerebral hypoperfusion white matter damage model by BCAS, while there was no stenosis in sham group. 7T Magnetic Resonance Imaging, Luxol Fast Blue(LFB) staining and electron microscope were used to observe and calculate the white matter lesion. The cognitive function was estimated by 8-arm maze task. Results: There was a significant change in DTI correlated parameter FA and RD, indicating glia-axonal integrity management and demyelination changes of white matter regions in BCAS group(P0.05). However, no changes in AD value indicating axonal damage was observed. LFB staining and electron microscope also demonstrated a significant demyelinating change in white matter regions(P0.05). In the working memory tasks test with the 8-arm radial maze, the BCAS mice made significantly more errors than the control mice(P0.001). Again, there were no detectable differences in the reference memory tasks between the 2 groups. There existed significant correlation between white matter lesion demonstrated by DTI and working memory errors(P0.05). Conclusion: At day 30 after BCAS, working memory deficits accompanying white matter changes were apparent in the mice, and the working memory deficit is attributable to damage of the frontal-subcortical circuits.
