Effects of “Nourishing Liver and Kidney” Acupuncture Therapy on Expressions of Vascular Endothelial Growth Factor and Synaptophysin after Cerebral Ischemia Reperfusion in Rats

Objective: To investigate the effects of acupuncture therapy on expressions of vasular endothelial growth factor(VEGF)and synaptophysin(SYN)after cerebral ischemia reperfusion in rats. Methods: One hundred and forty-four healthy adult male SD rats were randomly divided into sham operation group(n=30), model group(n=42), acupuncture group(n=42) and acupuncture control group(n=30). The middle cerebral ischemia reperfusion model(MCAO) was established by suture method. Acupunctures were performed in the acupuncture group and acupuncture control group with at main acupoints of Taixi(K103), Taichong(ST09) of both sides, for30 min. Then acupunctures were followed once daily in every morning, every 7-day for a course(acupuncture 6days and rest 1 day). The sham operation group, model group were conventionally fed in the cages, without any intervention therapy. The rats of each group were assessed with Modified neurological severity(m NSS) scores,the infarct volume was calculated by TTC staining and the expressions of VEGF and SYN were detected with the method of immunohistochemical SP at the 3thd, the 7thd and 14 th d. Results: Rats in acupuncture control group and sham operation group were presented no neurological deficit. The nerve functional recovery of rats in acupuncture group were significantly better than those in model group at the 7thd and 14 thd after modeling. Acupuncture control group and sham operation group were presented no the infarct volume. The infarct volumes of acupuncture group were significantly smaller than those in model group at the 7thd and 14 thd. At the each time point,the expressions of VEGF and SYN around the cerebral ischemia regions increased significantly in the model group, compared with the sham operation group and the acupuncture control group. In acupuncture group the expressions of VEGF at the 7thd, the 14 thd increased more significantly than those in the model group. In acupuncture group the expressions of SYN at the each time point increased more significantly than those in the model group. Conclusion: The acupuncture therapy can promote the recovery of neurological function of focal cerebral ischemia reperfusion in rats which may be associated with increase of VEGF and SYN in the infarct regions.
