Effect of dendritic cells pulsed with Tα146-162 on the expression of Crry, CD59a, and CD59b mRNA in experimental autoimmune myastheia gravis

Objective To study the expression of Crry, CD59a and CD59b mRNA in the muscle of mice immunized with T-AChR and of the immunized mice injected with immature myeloid dendritic cells (iMDCs) pulsed with immunodominant peptide T-AChRα146-162 (Tα146-162), and to investigate the effect of iMDC pulsed with Tα146-162 (Tα146-162- iMDC) induced antigen specific tolerance and the effect of the tolerance on the expression of Crry, CD59a and CD59b mRNA. Methods Precursor cells from mice bone marrow were cultured and differentiated in medium supplemented with GM-CSF and IL-10 for six days. Immature myeloid DCs were collected and pulsed with Tα146-162. 30 C57BL/6 mice were randomly divided into model group (A, n=12), intervention group (B, n=12) and normal control group (C, n=6). Mice of group A and B were immunized with T-AChR in CFA. Mice of group B were subcutaneously injected with Tα146-162-iMDC. Mice of group A and C received injections of PBS. Mice were evaluated for Christadoss standard, lymphocyte proliferation test was used to confirm the onset of experimental autoimmune myastheia gravis (EAMG). On the day 90 after the first immunization, mice were sacrificed, lymphocyte proliferation was assayed and Crry, CD59a and CD59b mRNA were detected by RT-PCR method. Results The average clinical scores of group A and group B mice were 1.67±1.15 vs 0.33±0.65 (P0.01), and the accumulated incidences were 75% vs 25% (P0.05). Compared with group A mice, group B mice had a significantly reduced lymphocyte proliferative response (P0.01) . The expression of Crry mRNA and CD59a mRNA decreased significantly in muscle of group A mice compared with that of group C mice (P0.05). The decrease was inhibited in mice of group B injected with Tα146-162-iMDC, but the expression of Crry mRNA was still lower than that of group C mice (P0.05). There was no significant difference in CD59a mRNA expression in muscle between group B and C (P0.05). CD59b mRNA could not be detected in the muscle of three groups. Conclusions Tα146-162-iMDC could prevent EAMG. The decrease of Crry mRNA and CD59a mRNA may involve in the pathogenesis of EAMG but CD59b could not correspond with it. The protective effect of Tα146-162-iMDC could be associated with the increased expression of Crry mRNA, CD59a mRNA in muscle and the antigenic specific suppression of lymphocyte proliferation.
