Correlation of the polymorphisms of apolipoprotein E gene and low-density lipoprotein receptor-related protein gene with sporadic Alzheimer’s disease

Objective To explore the roles of the polymorphisms of apolipoprotein E (ApoE) gene and exon3 polymorphism of low-density lipoprotein receptor-related protein (LRP) gene in the development of sporadic Alzheimer’s disease (SAD). Methods The polymorphisms of ApoE gene and exon3 polymorphism of LRP gene were detected by the polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) technique in 56 patients with SAD and 100 normal controls. Results The frequency of allele ε4 in patients with SAD increased significantly compared with that of the normal controls (x2=11.36, P0.01). SAD was positively correlated with allele ε4 (OR=3.29, CI 1.36-7.95) and LRP allele C (OR=1.83, P0.05), but was negatively associated with LRP allele T (OR=0.55, P0.05). Conclusions The allele ε4 of ApoE gene and allele C of LRP gene might be the risk factors for SAD. There may be a genetic association between the polymorphisms of ApoE gene and LRP gene and SAD.
